Hello,

i have oracle vm virtualbox 4.0. i ask if i do the update to 4.3, did i
lost the virtual machines (the instances) already installed or not ?

No updating or uninstalling VirtualBox does not uninstall/remove your
VMs. If you are updating from a really old version, it may be wise to
take a backup of your VMs.
